Laboratory: Microscopic species will be sorted out and mounted under binocular and compound microscopes in the laboratory at Gump. Macroalgae requiring critical microscopic taxonomic identification using internal anatomy will be sampled and analyzed in the laboratory, and extensive use made of available algal taxonomic literature (N’Yeurt reprint collection andbooks), UPF herbarium specimens, and online databases (AlgaeBase). Microphotographs will be taken using a digital camera mounted on a microscope. Voucher slide specimens will be produced and deposited in UPF herbarium. Tissue samples will be provided for DNA extraction in Moorea and sequencing will be carried out at SI as for other Plants (see 1.1.6)
Timeline:We envision three field scientists for 75 days of collection, documentation and vouchering. A post-doctoral researcher (Dr. Antoine D. R. N’Yeurt) will be employed to process the samples for taxonomical identification and for targeted barcode sequences based on protocols established in collaboration with CBOL (Saunders, Alison, etc.).
